MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA-c) is a 16‑amino‑acid mitochondrial‑derived peptide encoded by mitochondrial DNA, first identified in 2015. It acts as a key metabolic signaling molecule that translocates to the nucleus under stress, activates the AMPK pathway via inhibiting the folate cycle and promoting AICAR accumulation, enhances glucose uptake, fatty acid oxidation, and mitochondrial biogenesis, improves insulin sensitivity, reduces fat storage, suppresses inflammation and oxidative stress, and supports anti‑aging, metabolic health, and exercise performance; it is typically administered subcutaneously at 5–10 mg daily or 2–3 times weekly, with mild side effects and no significant impact on pituitary hormones.
